The Tn antigen is a type of tumor-associated carbohydrate antigen (TACA) that is expressed on the surface of most types of cancer cells, including breast, ovarian, pancreatic, and colorectal cancers. It is characterized by the presence of an abnormal terminal N-acetylgalactosamine (GalNAc) sugar linked to the serine or threonine residues of glycoproteins. Usually, the Tn antigen is not expressed in healthy adult tissues, as the GalNAc residues are usually further glycosylated with polysaccharide chains. Exposing the immune system to the Tn antigen (or other TACA) initiates an immune cascade that can potentially culminate in the elimination of tumor cells presenting the targeted epitope. However, exposure to Tn antigen alone is not usually sufficient to evoke an immune response. Glycoconjugates, where the Tn antigen is attached to the appropriate immunogenic carrier molecules, are usually required to raise antibodies. For example, a three-component vaccine (MPLA–Tn–KRN7000, structure shown in the post) containing a TLR4 ligand (MPLA) and an iNKT cell agonist (KRN7000) induced the production of antibodies that could specifically recognize, bind, and lyse human Tn-positive cancer cells. The conjugates improved the survival rate and prolonged survival time of tumor-challenged mice through elicitation of high levels of IgG antibodies. These can be cells that are presenting pathogenic antigens that have been ingested (phagocytes or B-cells), cells infected by viruses that are presenting viral proteins or abnormal body cells that are presenting non-self antigens Inactive T-cells that have not yet encountered their specific antigen are continuously circulating in the blood. Once an inactive T-cell does encounter an APC presenting the specific antigen to which it is complementary, the T-cell receptors bind to the antigen, leading to T-cell activation The activated T-cell then divides rapidly by mitosis, producing many clones of this specific T-cell. The large number of T-cells produced by mitosis are then allocated one of three roles: - T-helper cells (CD4+ T-cells): these cells help to speed up the immune response by releasing tiny molecules (cytokines) that call upon other immune cells, such as phagocytes and B-cells, to help - Cytotoxic T-cells (CD8+ T-cells): these cells have the ability to physically destroy infected cells by initiating cell apoptosis. ☢️RADIOLABELING OF MONOCLONAL ANTIBODIES☢️ is a method used to utilise monoclonal antibodies (previous post) as a treatment for cancer In this process, monoclonal antibodies are tagged with radioactive isotopes through a process called radiolabeling, which involves chemically attaching a radioactive isotope to the antibody molecule. Remember, this antibody has been isolated and cultured to be complementary to a target antigen (for example, the antigen presented by a cancerous liver cell) Once labeled, these monoclonal antibodies are administered to cancer patients, where they seek out and bind to specific antigens present on the surface of tumour cells. Once an antigen-antibody complex is formed, the radioactive isotopes emit radiation, which damages the DNA of the cancer cells and ultimately destroys them Because the monoclonal antibody binding sites are complementary only to one antigen, the radiation therapy is delivered directly to the tumour cells, sparing healthy tissues from damage.
